when and how often to store eucentric focus
Added by jinghua tang over 12 years ago
It was stated "These plus a record of eucentric focus value are
required for the z-height correction to eucentric height." Could
you explain when and where in leginon do I store eucentric focus?
If I move away from the center of the gird or with bent grid, do
I need manually set eucentric height often?
Thank you!

I started a wiki for defining eucentric focus. See Eucentric focus.
Eucentric focus value saved is the electric current applied to microscope objective lens. It may change if the alignment is changed. You should at least store it once a month if scope high tension and other upstream parameters not dramatically changed. I teach everyone to do it at the start of the leginon setup, whether it is a one-day session or multiple days since it is critical enough in autofocusing. This way, one or more of them would remember to do so in a month....
Places to save eucentric focus:
- See Autofocus_Calibration_with_Beam_Tilt_Node Rotation Center Storage and Retrieval section.
- Also available in any focuser note (Z Focus, Focus, Tomography Focus etc.) in the dialogue window pops up when you are doing manual focusing.
