How can I use defocused diffraction rather than LM for getting images at very low magnification? I want to avoid turning off the objective lens. I tried getting a diff setting from the microscope and got an "invalid magnification" error from the program. Thanks.

You can not. None of the pyscope functions (the control and retrieval of the parameters regarding microscope) handles diffraction mode operation. It is not something that I can take on at this point.
