Best sq preset for JEM 3200s

Added by Anchi Cheng over 9 years ago

It is time for me to ask a question:

I am having trouble finding a good sq preset on NYSBC 3200FSC that is coupled with DE-20. Here are what I tried:

1. Low Mag Mode 600x with 20 um under focus. Resulting pixel size is 15.5 nm
Pro: See the whole 300 mesh square and contrast good enough to see hole edge on a negatively stained carbon-over-hole c-flat grid.
Con: Strong hysteresis in image-shift.

2. Mag1 Mode 2000x with a larger PL2 value and 20 um under focus (in free-lens control) that reduces the magnification seen by the camera.
Pro: See the whole square and image-shift seems more stable.
Con: No contrast.

3. Mag1 Mode 2000x with 1.2 mm over-focus. Resulting pixel size is 13 nm
Pro: See the whole 300 mesh square and contrast good enough to see hole edge on a negatively stained carbon-over-hole c-flat grid.
Con: Significant but maybe a bit better than case 1 image-shift hysteresis. Image is over-focused.

Has anyone found a better way to do this ?
