leginon on jeol with motorized multispecimen holder
Added by Anonymous almost 15 years ago
I intend to install Leginon on a Jeol 1230 for automated imaging of large montages across serial sections. Jeol now told me that they procuced a new motorized pentaholder (it is a purpose-build item and not yet in their official price list), which allows motor driven switching from one grid to the next. This of coures sounds fantastic for all kind of automatisation across several grids. Did possibly somebody already use this holder together with Leginon?

It should certainly be possible to integrate a multi-holder into Leginon. We have done multiple grid data collection in Leginon using our custom built robot which loads a series of grids from a storage box into a single grid holder, then inserts each into the scope. We have also used this same application for multiple grid data collection when there is no robot, and a human must do the grid changes. Using a multiple grid holder would be a hybrid technique where some grid changes are automatic, and human interaction is necessary between each set of grids. I think it should be fairly simple to modify Leginon to accomplish this.
I do not know details of how the other groups (LBL, NYSBC) got Leginon to work on their Jeol, but I imagine this new holder would simply add some new functions to the scripting interface to the microscope, and therefore new functions in the pyScope Jeol module. Or, if it is a completely independent device and software driver, then a Python interface would need to be developed.
