I found the condition used to decide target in hole
targeting and the condition for focus are different. So I found some cases
is one hole is not a collection target but is a focus target. What will
happen?

The same thing happen at exposure targeting. I found some hole is a
collection target but does not have focus target. Is that OK? (these are
default) Should I make them the same?

Hole_Targeting autofocus normally get many targets fitted in the lattice (Magenta targets). It is up to you how you want the focus target to be selected from these Magenta targets. Just try the three options on the same square without submitting them, then you will see how they behave differently. People use "Good Hole" option when their are many holes that can be passed to be an acquisition target (green). Use "Any hole" when there is a fair chance that most lattice targets will fail the criteria to become an acquisition target.

It is not. Each image with acquisition target should have focus target accompany them or there is not focusing done and you final image is likely out-of-focus. Exposure_Targeting node uses Focus Template and then use the Focus Template Thickness parameters to pick one of focus target on the template. You should adjust the Focus Template Thickness parameters so that any reasonable acquisition (collection as you call it) target will find a focus target. Focus target just need to avoid very thick ice, grid bar, and broken carbon film, so the criteria should be looser
than acquisition.
