4 | |||
5 | Dose Calibration is a calibration of CCD sensitivity. This calibration is required only |
||
6 | once at each high tension per instrument. It can be used for three purposes: |
||
7 | |||
8 | |||
9 | |||
10 | |||
11 | * To obtain a scaling factor between the scope screen current and the beam current |
||
12 | measured by a standardized equipment such as a Faraday cup. |
||
13 | |||
14 | |||
15 | * To calibrate the camera sensitivity to electron. |
||
16 | |||
17 | |||
18 | * If camera sensitivity is already calibrated, the value can be entered |
||
19 | directly. |
||
20 | |||
21 | |||
22 | |||
23 | |||
24 | |||
25 | |||
26 | |||
27 | h2. Determine the screen current/beam current scale factor |
||
28 | |||
29 | |||
30 | |||
31 | The following uses a Faraday cup |
||
32 | |||
33 | |||
34 | |||
35 | |||
36 | * Scope> Move the stage to an empty area or a broken hole. |
||
37 | |||
38 | |||
39 | * Scope> Adjust the beam so that it passes through the measuring area of the |
||
40 | Faraday cup. |
||
41 | |||
42 | |||
43 | * Scope> Choose a magnification so that the same beam will pass within the main |
||
44 | screen, too. |
||
45 | |||
46 | |||
47 | * Leginon/Dose/Dose Calibrator> Beam Diameter = 0.16 m for Tecnai main |
||
48 | screen. |
||
49 | |||
50 | |||
51 | * Leginon/Dose/Dose Calibrator> Enter Screen Current to Beam Current Scale |
||
52 | Factor as 1 as a starting point. |
||
53 | |||
54 | |||
55 | * scope> Optionally lower the screen. Dose Calibrator will automatically do this |
||
56 | if the user forgets. |
||
57 | |||
58 | |||
59 | * Leginon/Dose/Dose Calibrator> Click Measure Dose Rate. The Dose Rate, Beam |
||
60 | Current, and Screen Magnification will appear in the corresponding fields after they |
||
61 | have been calculated. |
||
62 | |||
63 | |||
64 | * Leginon/Dose/Dose Calibrator> Adjust the Screen Current to Beam Current Scale |
||
65 | Factor so that the displayed beam current is equal to that of the Faraday cup |
||
66 | measurement. NRAMM Tecnai's value is 0.88. |
||
67 | |||
68 | |||
69 | |||

72 | |||
73 | |||
74 | |||
75 | |||
76 | h2. Calibrate the camera sensitivity |
||
77 | |||
78 | |||
79 | |||
80 | |||
81 | |||
82 | * Scope> Move the stage to an empty area or a broken hole. |
||
83 | |||
84 | |||
85 | * Scope> Expand the beam so that it covers the main screen and illuminate it |
||
86 | uniformly. |
||
87 | |||
88 | |||
89 | * Leginon/Dose/Settings> Configure camera with overide preset activated. |
||
90 | |||
91 | |||
92 | * Leginon/Dose/Dose Calibrator> |
||
93 | |||
94 | |||
95 | |||
96 | Beam Diameter = 0.16 m for the Tecnai's main viewing screen. |
||
97 | |||
98 | |||
99 | Screen Current to Beam Current Scale Factor = (as determined above or 1 to assume |
||
100 | good calibration by the microscope manufacturer) |
||
101 | |||
102 | |||
103 | * scope> Optionally lower the screen. Dose Calibrator will automatically do this |
||
104 | if the user forgets. |
||
105 | |||
106 | |||
107 | * Leginon/Dose/Dose Calibrator> Click Measure Dose Rate. The Dose Rate, Beam |
||
108 | Current, and Screen Magnification will appear in the corresponding fields after they |
||
109 | have been calculated. |
||
110 | |||
111 | |||
112 | * Leginon/Dose/Dose Calibrator> Without changing the beam intensity, click on |
||
113 | "Calibrate". An image is acquired by Leginon. |
||
114 | |||
115 | |||
116 | *Leginon will automatically lift the main viewing screen on the microscope. Some |
||
117 | users report that the function does not work on their microscopes. Do it manually if |
||
118 | the automatic function does not work. |
||
119 | |||
120 | |||
121 | * The calculated CCD sensitivity (counts / electron) will display once the |
||
122 | calculation is completed. |

133 | |||
134 | |||
135 | |||
136 | |||
137 | |||
138 | * Leginon/Dose/Dose Settings/Camera Sensitivity> Enter Sensitivity values in |
