h1. Acquire images on Falcon II h2. Preparation in TUI/TIA h3. Acquire Gain/Dark References Follow FEI's instruction in obtaining Gain/Dark References. If you plan to save frames, make sure you repeat the process with "Store intermediate images" option on. In addition, take your Gain/Dark Reference at the dose rate of your data collection when using this option. The linearization feature in Falcon software does not work when "Store intermediate images" is on. Take an image at a long exposure time after reference image acquisition to confirm that the references are indeed valid in this case. h2. Recommended dose rate The optimal dose rate is likely between 3 - 3.5 electrons/pixel/frame or 50-60 electron/pixel/s based on available literature of high resolution structures using this camera.
